Calculation of droplet deformation at intermediate Reynolds number using a Volume of Fluid technique
The deformation of a droplet or bubble in simple extensional flow fields is investigated at intermediate Reynolds number using a Volume-of-Fluid (VOF) numerical algorithm (MAC2) developed by Rudman. The results of an evaluation study of the MAC2 algorithm for such flows are presented. The final steady shapes of droplets in axisymmetric extensional flow fields determined by MAC2 are compared with the published numerical results of Ramaswamy and Leal. This work forms the initial stage of a more comprehensive study of droplet deformation in time-dependent shear fields experienced by drops flowing through devices (e.g.\ homogenisers) designed for droplet breakup
A Cohort Study of Traffic-Related Air Pollution and Mortality in Toronto, Ontario, Canada
BackgroundChronic exposure to traffic-related air pollution (TRAP) may contribute to premature mortality, but few studies to date have addressed this topic.ObjectivesIn this study we assessed the association between TRAP and mortality in Toronto, Ontario, Canada.MethodsWe collected nitrogen dioxide samples over two seasons using duplicate two-sided Ogawa passive diffusion samplers at 143 locations across Toronto. We calibrated land use regressions to predict NO2 exposure on a fine scale within Toronto. We used interpolations to predict levels of particulate matter with aerodynamic diameter < or = 2.5 microm (PM(2.5)) and ozone levels. We assigned predicted pollution exposures to 2,360 subjects from a respiratory clinic, and abstracted health data on these subjects from medical billings, lung function tests, and diagnoses by pulmonologists. We tracked mortality between 1992 and 2002. We used standard and multilevel Cox proportional hazard models to test associations between air pollution and mortality.ResultsAfter controlling for age, sex, lung function, obesity, smoking, and neighborhood deprivation, we observed a 17% increase in all-cause mortality and a 40% increase in circulatory mortality from an exposure contrast across the interquartile range of 4 ppb NO2. We observed no significant associations with other pollutants.ConclusionsExposure to TRAP was significantly associated with increased all-cause and circulatory mortality in this cohort. A high prevalence of cardiopulmonary disease in the cohort probably limits inference of the findings to populations with a substantial proportion of susceptible individuals
APRIL is a novel clinical chemo-resistance biomarker in colorectal adenocarcinoma identified by gene expression profiling
<p>Abstract</p> <p>Background</p> <p>5-Fluorouracil(5FU) and oral analogues, such as capecitabine, remain one of the most useful agents for the treatment of colorectal adenocarcinoma. Low toxicity and convenience of administration facilitate use, however clinical resistance is a major limitation. Investigation has failed to fully explain the molecular mechanisms of resistance and no clinically useful predictive biomarkers for 5FU resistance have been identified. We investigated the molecular mechanisms of clinical 5FU resistance in colorectal adenocarcinoma patients in a prospective biomarker discovery project utilising gene expression profiling. The aim was to identify novel 5FU resistance mechanisms and qualify these as candidate biomarkers and therapeutic targets.</p> <p>Methods</p> <p>Putative treatment specific gene expression changes were identified in a transcriptomics study of rectal adenocarcinomas, biopsied and profiled before and after pre-operative short-course radiotherapy or 5FU based chemo-radiotherapy, using microarrays. Tumour from untreated controls at diagnosis and resection identified treatment-independent gene expression changes. Candidate 5FU chemo-resistant genes were identified by comparison of gene expression data sets from these clinical specimens with gene expression signatures from our previous studies of colorectal cancer cell lines, where parental and daughter lines resistant to 5FU were compared. A colorectal adenocarcinoma tissue microarray (n = 234, resected tumours) was used as an independent set to qualify candidates thus identified.</p> <p>Results</p> <p>APRIL/TNFSF13 mRNA was significantly upregulated following 5FU based concurrent chemo-radiotherapy and in 5FU resistant colorectal adenocarcinoma cell lines but not in radiotherapy alone treated colorectal adenocarcinomas. Consistent withAPRIL's known function as an autocrine or paracrine secreted molecule, stromal but not tumour cell protein expression by immunohistochemistry was correlated with poor prognosis (p = 0.019) in the independent set. Stratified analysis revealed that protein expression of APRIL in the tumour stroma is associated with survival in adjuvant 5FU treated patients only (n = 103, p < 0.001), and is independently predictive of lack of clinical benefit from adjuvant 5FU [HR 6.25 (95%CI 1.48-26.32), p = 0.013].</p> <p>Conclusions</p> <p>A combined investigative model, analysing the transcriptional response in clinical tumour specimens and cancers cell lines, has identified APRIL, a novel chemo-resistance biomarker with independent predictive impact in 5FU-treated CRC patients, that may represent a target for novel therapeutics.</p
Green Plants in the Red: A Baseline Global Assessment for the IUCN Sampled Red List Index for Plants
Plants provide fundamental support systems for life on Earth and are the basis for all terrestrial ecosystems; a decline in plant diversity will be detrimental to all other groups of organisms including humans. Decline in plant diversity has been hard to quantify, due to the huge numbers of known and yet to be discovered species and the lack of an adequate baseline assessment of extinction risk against which to track changes. The biodiversity of many remote parts of the world remains poorly known, and the rate of new assessments of extinction risk for individual plant species approximates the rate at which new plant species are described. Thus the question ‘How threatened are plants?’ is still very difficult to answer accurately. While completing assessments for each species of plant remains a distant prospect, by assessing a randomly selected sample of species the Sampled Red List Index for Plants gives, for the first time, an accurate view of how threatened plants are across the world. It represents the first key phase of ongoing efforts to monitor the status of the world’s plants. More than 20% of plant species assessed are threatened with extinction, and the habitat with the most threatened species is overwhelmingly tropical rain forest, where the greatest threat to plants is anthropogenic habitat conversion, for arable and livestock agriculture, and harvesting of natural resources. Gymnosperms (e.g. conifers and cycads) are the most threatened group, while a third of plant species included in this study have yet to receive an assessment or are so poorly known that we cannot yet ascertain whether they are threatened or not. This study provides a baseline assessment from which trends in the status of plant biodiversity can be measured and periodically reassessed
Beta defensin-2 is reduced in central but not in distal airways of smoker COPD patients
Background: Altered pulmonary defenses in chronic obstructive pulmonary disease (COPD) may promote distal airways bacterial colonization. The expression/activation of Toll Like receptors (TLR) and beta 2 defensin (HBD2) release by epithelial cells crucially affect pulmonary defence mechanisms. Methods: The epithelial expression of TLR4 and of HBD2 was assessed in surgical specimens from current smokers COPD (s-COPD; n = 17), ex-smokers COPD (ex-s-COPD; n = 8), smokers without COPD (S; n = 12), and from non-smoker non-COPD subjects (C; n = 13). Results: In distal airways, s-COPD highly expressed TLR4 and HBD2. In central airways, S and s-COPD showed increased TLR4 expression. Lower HBD2 expression was observed in central airways of s-COPD when compared to S and to ex-s-COPD. s-COPD had a reduced HBD2 gene expression as demonstrated by real-time PCR on micro-dissected bronchial epithelial cells. Furthermore, HBD2 expression positively correlated with FEV1/FVC ratio and inversely correlated with the cigarette smoke exposure. In a bronchial epithelial cell line (16 HBE) IL-1β significantly induced the HBD2 mRNA expression and cigarette smoke extracts significantly counteracted this IL-1 mediated effect reducing both the activation of NFkB pathway and the interaction between NFkB and HBD2 promoter. Conclusions: This study provides new insights on the possible mechanisms involved in the alteration of innate immunity mechanisms in COPD. © 2012 Pace et al
Longitudinal change in the BODE index predicts mortality in severe emphysema
Rationale: The predictive value of longitudinal change in BODE (Body
mass index, airflow Obstruction, Dyspnea, and Exercise capacity)
index has received limited attention. We hypothesized that decrease
in a modified BODE (mBODE) would predict survival in National
Emphysema Treatment Trial (NETT) patients.
Objectives: To determine how the mBODE score changes in patients
with lung volume reduction surgery versus medical therapy and correlations
with survival.
Methods: Clinical data were recorded using standardized instruments.
The mBODE was calculated and patient-specific mBODE trajectories
during 6, 12, and 24 months of follow-up were estimated using
separate regressions for each patient. Patients were classified as
having decreasing, stable, increasing, or missing mBODE based on
their absolute change from baseline. The predictive ability of mBODE
change on survival was assessed using multivariate Cox regression
models. The index of concordance was used to directly compare the
predictive ability of mBODE and its separate components.
Measurements and Main Results: The entire cohort (610 treated medically
and 608 treated surgically) was characterized by severe airflow
obstruction, moderate breathlessness, and increased mBODE at baseline.
A wide distribution of change in mBODE was seen at follow-up. An
increase in mBODE of more than 1 point was associated with increased
mortality in surgically and medically treated patients. Surgically
treated patients were less likely to experience death or an increase
greater than 1 in mBODE. Indices of concordance showed that mBODE

Enrichment measurement by passive γ-ray spectrometry of uranium dioxide fuel pellets using a europium-doped, strontium iodide scintillator
The performance of a europium-doped strontium iodide scintillator for uranium enrichment measurement of a variety of sintered uranium dioxide fuel pellets is described and compared to that of caesium iodide and sodium iodide. Enrichment has been determined via passive γ-ray spectrometry of the 186 keV line from uranium-235 using gross count, net count, and peak ratio analyses. The 38 mm Ø x 38 mm strontium iodide crystal demonstrates superior energy resolution (3.43 ± 0.03% at 662 keV) and competitive detection efficiency for its size in the energy range of interest for uranium enrichment analysis (<250 keV). It demonstrates better χ v 2 and coefficient of determination values than caesium iodide and sodium iodide when measuring uranium enrichment using the gross- and net-count from the 186 keV emission. It is shown to have the least measurement variance of the three scintillators studied in determining the uranium enrichment of pellets in a blind test, with a relative error comparative to sodium iodide and smaller than caesium iodide. This research heralds the potential of strontium iodide in passive γ-ray uranium enrichment applications
Discovery of Inhibitors of Leishmania β-1,2-Mannosyltransferases Using a Click-Chemistry-Derived Guanosine Monophosphate Library
Leishmania spp. are a medically important group of protozoan parasites that synthesize a novel intracellular carbohydrate reserve polymer termed mannogen. Mannogen is a soluble homopolymer of β-1,2-linked mannose residues that accumulates in the major pathogenic stages in the sandfly vector and mammalian host. While several steps in mannogen biosynthesis have been defined, none of the enzymes have been isolated or characterized. We report the development of a simple assay for the GDP-mannose–dependent β-1,2-mannosyltransferases involved in mannogen synthesis. This assay utilizes octyl α-d-mannopyranoside to prime the formation of short mannogen oligomers up to 5 mannose residues. This assay was used to screen a focussed library of 44 GMP-triazole adducts for inhibitors. Several compounds provided effective inhibition of mannogen β-1,2-mannosyltransferases in a cell-free membrane preparation. This assay and inhibitor compounds will be useful for dissecting the role of different mannosyltransferases in regulating de novo biosynthesis and elongation reactions in mannogen metabolism
